In addition, we investigated the potential of using Viagra or cyclic GMP in the treatment of erectile dysfunction in a human model of erectile dysfunction.
A total of 18 male Wistar rats (180-250 g) were used in this study and they were randomly divided into 4 groups (n=10): (1) control group (CON) (CON) - animals that received a placebo (CON) - normal saline (NS) - saline (NS) - Viagra (Viagra) (100 mg/ml) - vehicle (NS) - vehicle (NS) - vehicle (NS). The animals were housed in a temperature (22-25 °C) and humidity (50-80%) controlled environment with a 12 h/12 h light/dark cycle. All experimental procedures were in accordance with the guidelines of the Laboratory Animal Use Committee of the National Institutes of Health.
At the end of the experimental period, the rats were anesthetized with 1% pentobarbital (N=10) or 0.1% pentobarbital (N=10) (Euthy-Teva) and the right hind legs were dissected. The penis was removed, washed, and weighed. To a total of 10 μl of each rat’s blood were withdrawn from the corpus cavernosum using a sterile EDTA needle. The plasma and whole blood plasma were separated by centrifugation at 3000 rpm for 10 min at 4 °C. The plasma were extracted with a 100 μl Eppendorf tubes and stored at −80 °C until analysis.
To investigate the effects of Viagra on the phosphodiesterase type-5 (PDE-5) activity of the human corpus cavernosum (hC) isolated from rat, the effects of Viagra on hC in vitro were examined by means of the phosphodiesterase type-5 (PDE-5) inhibitor pdi-test. The results were expressed as the mean of three independent experiments. The inhibition of PDE-5 by Viagra was confirmed by the inhibition of the inhibition of the PDE-5 inhibitor activity of the human corpus cavernosum (hC) isolated from rat. The data were analyzed by one-way ANOVA and Tukey’s multiple comparisons test. In addition, the statistical analysis was conducted by use of the software SPSS v.18.0 (SPSS, Chicago, IL, USA).P<0.05 was considered significant.

The effects of Viagra and cyclic GMP on hC isolated from rat were examined by means of the phosphodiesterase type-5 (PDE-5) inhibitor pdi-test.
